I don't use photoshop, maybe one of the designers that do will be able to help.
However, I get the feeling you have misunderstood the instructions.
1) open the mesh in photoshop
2) add a transparent layer to the file (not a duplicate of the mesh)
3) use the lasso tool to mark the outline you want
4) bucket fill on the tranparent layer
5) delete any areas you do not want
6) save as a photoshop file using the nameing convention described in the how to's
7) hide the mesh layer
8) reduce the image to a size for publishing (usually 256x256 or 512x512)
9) save as a .png file
Make sure you get to grips with your graphics program, there are loads of tutorials on the net for basic operations.
